Alxyl
Alxyl is sequence-independent modification, which can be used to add an Alkyne group via a C6 (hexynyl)- linker to the phosphate of the 5´-terminus of an oligonucleotide. With this modification, a sequence of 4 carbons atoms separate the oligonucleotide from the alkyne moiety, thereby reducing steric interactions between the reactive group and the oligonucleotide itself. In this context, multiple sites within the oligo can be labelled by clicking a variety of azide-containing labels/tags to different alkyn groups. To learn more about click-chemistry, click here.
